Uplistsikhe was first inhabited in the early Bronze Age and rose to prominence in the 1st millennium BC as a major religious and political center of the Kingdom of Iberia. The town reached its height in the 6th-5th centuries BC when it was a major spiritual center for the pagan Georgian pantheon. By the 3rd century BC, Uplistsikhe had declined in importance but regained some of its former statuses under King Pharnavaz I of Iberia in the 2nd century BC.

The town declined again by the end of the 1st century AD after a series of invasions from northern tribes. It was further abandoned after being sacked by the Mongols in 1259. The site was rediscovered in 1887 by Jakob Kutschinski, a Polish historian, who carried out excavations at Uplistsikhe from 1888 to 1891.
